NOD.Cg-Prkcdscid Il2rgtm1Wjl/SzJ (NSG) neonatal mice were sub-lethally irradiated and injected with 1 x 10^6 human fetal liver CD34+ cells per mouse. Nine humanized mice were infected with the HIV-1 strain NL4-3-HA (47) for 10 weeks, and then 4 were switched to ART-containing feed. Humanized mice between ages 12-20 weeks, engrafted with at least 30% human CD45+ cells in blood, of which at least 10% were CD4+, were infected via intraperitoneal injections containing 5-10 x 10^5 infectious units of HIV-1. This formulation was calculated by consideration of the average amount of food consumed by the mice (3.5g per day) and their average weight (20g) and provided average daily drug doses of Emtricitabine (FTC) at 87.5mg/kg/day, Tenofovir (TDF) at 131mg/kg/day, and Raltegravir (RAL) and Darunavir (DRV) at 175mg/kg/day each. |
