Developing a BLT model on the C57BL/6 background C57BL/6 Rag22/2gc2/2 mice have been described previously.23-25 CD47-null B6.129-CD47tm1Fpl/J mice (The Jackson Laboratory, Bar Harbor,ME) were crossed with C57BL/6 Rag22/2gc2/2 females, and F1 Rag21/2gc-/Y CD471/2 males were backcrossed with Rag22/2gc2/2 females. Mating of F2 Rag22/2gc2/2CD471/2 females and Rag22/2gc -/YCD471/2 males produced
theRag22/2gc2/2CD472/2 (TKO) strain.Forty-eight TKO-BLT mice were inoculated intrarectally with 10^5 TCIU of HIVJR-CSF. Twenty-six (54%) of the mice became infected with p24 plasma titers peaking between 2 and 4 weeks postinfection (wpi) and then declining over the next several weeks. |
