Briefly, preconditioned NOD/SCID-gamma chain null (NSG) mice (The Jackson Laboratories) implanted with thymus and liver tissue (Advanced Bioscience Resources) were transplanted with 2×10^5 to 3×10^5 autologous human fetal liver CD34+ cells and monitored for human engraftment.Humanized BLT mice are susceptible to vaginal, rectal, and intravenous HIV infection (10– 12, 36). We infected BLT mice with CCR5-tropic HIV (JR-CSF or SF162). Infected BLT mice received ART consisting of daily intraperitoneal injections of emtricitabine (FTC; 140 to 200 mg/kg body weight), tenofovir disoproxil fumarate (TDF; 146 to 208 mg/kg), and raltegravir (RAL; 56 to 80 mg/kg). BLT mouse that received ART for 58 days (followed for a total of 77 days postexposure) as well as in a pair of untreated, infected BLT mice (followed for a total of 69 and 70 days postexposure) |
