首先将PD-L1Flox/-小鼠和C57BL/6小鼠进行杂交，得到PD-L1Flox/-小鼠，然后将PD-L1Flox/-小鼠进行自交，得到PD-L1Flox/Flox小鼠，将雌性PD-L1Flox/Flox小鼠和带有巨噬细胞特异性表达 Cre 酶的Lyz2-iCre雄鼠进行杂交，得到 PD-L1Flox/- with Cre小鼠，最后将PD-L1Flox/Flox小鼠和PD-L1Flox/- with Cre小鼠杂交，得到1/4的PD-L1Flox/Flox with Cre小鼠，即为巨噬细胞特异性PD-L1敲除小鼠。 |
