制作方法Making method Six-week-old female hDPP4-Tg mice27 were maintained in a pathogen-free facility and housed in cages containing sterilized feed and drinking water.r. MERS-CoV (HCoVEMC2012 strain) was propagated and titrated on Vero cells. Following intraperitoneal anesthetization with sodium pentobarbital (5 mg/kg of body weight), mice were treated intravenously (600 μg/kg, i.v.) with a monoclonal Ab (mAb) against mouse C5aR (Hycult Biotech,The Netherlands) or phosphate-buffered saline (PBS) as a sham treatment control. All mice were then intranasally inoculated with MERS-CoV (103.3 50% tissue culture infectious dose (TCID50)) in 20 μl Dulbecco’s modified Eagle’s medium.
